There are many kinds of twists that don’t work. In the case of Splice, this was a twist that didn’t work because people wanted to throw up when they watched it (indeed, some people actually booed in the theater).
The plot centers around genetic engineers who experiment with combining humans with animals – and if you think that’s gross, you don’t want to know what ends up happening in the end. Let’s just say this is one twist you should be happy we’re not revealing it for you…
